چکیده

Rab8 (Ras-related proteins in brain 8) is a 24 kDa protein that belongs to the small Rab GTPase family (7). Rab proteins are known for their participation in and regulation of intracellular membrane trafficking pathways (17, 46, 48). In their active GTP-bound state different Rab proteins bind to different membrane compartments and recruit specific effector proteins, which are not only involved in docking and fusion with the target membrane but also in the formation of transport vesicles and in binding motor proteins for vesicle transport (17, 18, 28, 45, 48).

Rab8 directs furrow ingression and membrane addition during epithelial formation in Drosophila melanogaster.

One of the most fundamental changes in cell morphology is the ingression of a plasma membrane furrow. The Drosophila embryo undergoes several cycles of rapid furrow ingression during early development that culminate in the formation of an epithelial sheet. Previous studies have demonstrated the requirement for intracellular trafficking pathways in furrow ingression; however, the pathways that l...

FIP-2, a coiled-coil protein, links Huntingtin to Rab8 and modulates cellular morphogenesis

Huntington's disease is characterised by the death of cortical and striatal neurons, and is the result of an expanded polyglutamine tract in the Huntingtin protein [1]. Huntingtin is present on both endocytic and secretory membrane organelles but its function is unclear [2,3]. Rab GTPases regulate both of these transport pathways [4]. We have previously shown that Rab8 controls polarised membra...

Mutant rab8 Impairs docking and fusion of rhodopsin-bearing post-Golgi membranes and causes cell death of transgenic Xenopus rods.

Rab8 is a GTPase involved in membrane trafficking. In photoreceptor cells, rab8 is proposed to participate in the late stages of delivery of rhodopsin-containing post-Golgi membranes to the plasma membrane near the base of the connecting cilium. To test the function of rab8 in vivo, we generated transgenic Xenopus laevis expressing wild-type, constitutively active (Q67L), and dominant negative ...

GEF-mechanism of the Rab8-GEFs Rabin8/GRAB 1 Intermediates in the Guanine Nucleotide Exchange Reaction of Rab8 Catalyzed by Rabin8/GRAB

Small G-proteins (G) of the Ras-superfamily control the temporal and spatial coordination of intracellular signaling networks by acting as molecular on/off switches. Guanine nucleotide exchange factors (GEFs) regulate the activation of these G-proteins through catalytic replacement of guanosine diphosphate (GDP) by guanosine triphosphate (GTP). During nucleotide exchange, three distinct substra...

Caveolin-1 Induces Formation of Membrane Tubules That Sense Actomyosin Tension and Are Inhibited by Polymerase I and Transcript Release Factor/Cavin-1

Caveolin-1 and caveolae are often lost in cancer. We found that levels of caveolin-1 and polymerase I and transcript release factor (PTRF)/cavin-1 correlated closely in a panel of cancer and normal cells. Caveolin-1 reexpression in cancer cells lacking both proteins induced formation of long membrane tubules rarely seen in normal cells. PTRF/cavin-1 inhibited tubule formation when coexpressed w...
